Hamilton is a son of actor and director Martin Hamilton. At the age of thirteen he made his television debut in the drama series Cool from Teleac/NOT. 

A year later he played a role in The Secret of the Lindenborch, directed by his father. After this assignment, he appeared several times in SchoolTV television series. 

With his role as Fos Fischer in the soap opera Good Times, Bad Times, Hamilton had his big breakthrough on television. He played this role for 3 years and also obtained his HAVO diploma in between the duties. A year after his departure from the soap opera, he made his appearance in the youth series SpangaS, where he played the character Koen van Wageningen. In addition to the television series, he also had a role in the feature film SpangaS on Survival. 

In 2010 he started filming the popular drama series 'Overspel', in which he played the role of the adolescent Marco Steenhouwer for 3 seasons. In between, Hamilton also acts in the series 'Van God Los' and plays a role in the cinema film 'Manslaughter', in which he, as an aggressive street urchin, makes the life of ambulance brother, Theo Maassen, miserable. 

In 2015, Hamilton got the leading role in the feature film 'Fataal'. An intense story in which, as the owner of a nightclub, he himself becomes the victim of senseless violence. The film is still shown in schools to make students aware of the consequences of senseless violence.

After his role in Fataal, Hamilton plays various roles in, among others, 'Flikken Maastricht' and in the Telefilm 'Billy'. 

In 2020, Jeffrey appeared in Paul Ruven's documentary "Let's Dance". 

In 2021 he was asked for and played the role of a blind apostle in "The Passion 2021". 

In 2023 he was asked for the comedy series "Talk Powder" and he regularly plays various roles/characters. Jeffrey will also star in the series "Talk Powder" 2024.
